Significance of Nicotinic receptor binding

Nicotinic receptor binding was assessed using a radioligand binding assay. The procedure involved incubating protein samples at a concentration of 400 μg mL –1 with 0.75 nM of [3H]-cytisine, a nicotinic receptor ligand. Various concentrations of test compounds were added along with Tris-HCl buffer to a final volume of 1 mL. The mixture was incubated for 75 minutes at 4°C to facilitate binding interactions.
